  2. Completed assays: Lipids (total, HDL and LDL cholesterol and triglycerides) Glucose Creatinine Insulin (all MESA Family values with new assay) CRP (data on second half of MESA Family not released)

  3. Pending assays: • Multiplex panels for adipokines: • Adiponectin, resistin, leptin, TNF-α, MCP-1 • IL-6* (first half of MESA Family completed) • D-dimer* (first half of MESA Family completed) • PAI-1** • *Measured in full MESA cohort • **Measured in MESA 1000-person sample (Group 3)

  4. Tasks: • Complete remaining assay work: • IL-6 assays on second half of MESA Family participants using high sensitivity ELISA (carefully examine data; may need to re-run some assays) • D-dimer assays on second half of MESA Family participants • Adipokine panels – will be run on full MESA Family cohort including MESA Classic participants • PAI-1 – will be run on full MESA Family cohort including MESA Classic participants

IL-6 Assay Issue: • ~3 years ago, R&D Systems changed their high sensitivity IL-6 assay • New format potentially gives false high values (~10%) • Affects first half of MESA Family (MESA not affected) • No suitable replacement for R&D assay • After extensive work, it appears most values < 12 pg/ml are valid; approximately 50% of values ≥ 12 pg/ml are false high values • Complete MESA Family assays using the R&D Systems high sensitivity assay

  6. Options: • Use data “as is” – IL-6 and CRP highly correlated in first half of MESA Family participants • or • Re-measure values ≥ 12 pg/ml using another assay to identify false high values • Lab will cover the costs of the second assay • Caveat – second assay requires 250 μL of sample • In the first half of MESA Family, 17 IL-6 values ≥ 12 pg/ml; limited number of participants affected

  7. Time Frame: • In progress: • IL-6 assays • D-dimer assays • Started in several weeks: • Adipokine panels (adiponectin, resistin, leptin, TNF-α, MCP-1) • Pending: • PAI-1
