  4. Yeast Cells • Eukaryotic • Gene Sequences are well known • They are inexpensive to maintain • They reproduce at a quick rate • Cell response to DNA damage or other diseases is similar to humans.

Project Goals • To understand the yeast cell cycle by finding the amount of regulatory proteins that are present in different parts of the cell cycle • Develop a model to predict regulation of the Eukaryotic Cell cycle • Once understood, manipulate the cell environment to create a cancerous state. • Then determine ways to treat the cancerous cell.

  6. Polymerase Chain Reaction • (PCR) is a technique in molecular biology used to amplify a single or few copies of a piece of DNA.

  7. Polymerase Chain Reaction is comprised of 3 steps. Denaturation is the first regular cycling event consisting of heating the reaction. The Annealing stage lowers the temperature. During Elongation the DNA polymerase synthesizes a new DNA strand complementary to the DNA template strand.

  8. Gel Electrophoresis Is used for the separation of DNA,RNA, or protein molecules using an electric field applied to a gel matrix.

  9. Agarose Gel As a gel, an agarose medium is porous and therefore can be used to measure microorganism motility and mobility. Factors such as Conformations of a DNA plasmid, and length of the DNA molecule can determine how fast and how far a band moves. Ethidium Bromide is a common dye. It fluoresces under UV light when intercalated into DNA.
