Genetic frequency and HIV-1 High Risk Patients • Once CCR5 was identified as a co-receptor for HIV, researchers wanted to investigate if the genes for CCR5 affected resistance to HIV in cohorts • High risk patients were divided into infected and noninfected groups and a number of loci were examined to determine genetic frequencies.

Genetic Frequency and Loci • 170 loci were selected based on polymorphisms, involvement in retroviral infection (pathogenesis), and genetic location. • Test group: HIV-1-infected versus HIV-1-antibody-negative individuals • Only 1 of the loci, CCR5, displayed significant difference in distortion of genotype frequencies among test group. • Highly significant departure from genotype equilibrium in Caucasian homosexual men.

  4. A Distortion in CCR5 • Only CCR5 displayed significant difference in distortion of genotype frequencies among test group. • Because defect was found in only CCR5, expression of the defect may account for differences in susceptibility to HIV.
