  1. Marijuana Source and history of use Pharmacokinetics and dynamics Effects

  2. Cannabis sativa • Hemp, THC • Hashish, charas: resin, 10-20% THC • Ganja, sinsemilla: Dried tops, 5 - 8% THC • Marijuana, bhang: Remainder, 2 - 5 % THC

  3. Pharmacodynamics • Anandamide and the cannabinoid receptor • Inhibits hippocampal glutamate release • May inhibit potassium channel response • Cannabinoid receptors in basal ganglia, cerebellum, and cerebral cortex • No brainstem cannabinoid receptors, so vegetative functions are not affected directly.

  4. Pharmacokinetics • Smoke inhalation, eating, Marinol • High fat solubility • Elimination half-life: 30 hours to 4 days • Accumulation dosing • Positive urinalysis in heavy users for 1 month

  5. Effects • Depression and stimulation • Sedation and analgesia • Decrease voluntary movement: mellow • Lower aggression • Block convulsions and reflexes • Impair cognition • May cause hallucinations, time distortion

  6. More effects • Lower ovarian and possibly testicular function • High doses may produce anxiety reactions: 60% report at least one such experience • Impairs focus, attention in chronic users • Amotivational syndrome: Depression? • Immunosuppression • Gateway drug?
