How does a Mass Spectrometer work? • Separation in analyser according to mass-to-charge ratios (m/z) base on ionization • Detection of separated ions and their relative abundance
Fundamental parts 1) ion source - ions are produced, accelerated, and focused; 2) Analyzer - the beam is separated based on mass/charge ratios 3) Collector - the ion beams are measured either sequentially (single collector) or simultaneously (multi-collector).
3 major limitations: Compounds cannot be characterised without clean samples This technique has not the ability to provide sensitive and selective analysis of complex mixture For big molecules like peptides spectra are very complex and very difficult to interpret
