Quantitative detection of erlotinib-sensitizing EGFR kinase domain exon 19 in-frame deletion in resected NSCLC tumor genomic DNA using the nanofluidic digital array.Examples of digital-PCR array detection of EGFR exon 19 deletions in 10 patient genomic DNA samples (panels a-e, and g-k) are shown here, with 5,000 copies of DNA molecules each panel. Panel (f): 5 copies of Del 19 mutant DNA molecules from cell line HCC827 (as positive control) were spiked into 5,000 wild type EGFR genomic DNA molecules. Panel (l): 5,000 copies of wild type EGFR genomic DNA molecules (negative control). The tumor sample NSCLC-11 [DID-Panel (e)] showed one EGFR Del 19 allele (0.02%), not detectable by direct DNA sequencing. Part of this DID array is illustrated in Figure 3.
